How 5-Amino-1MQ Works: NNMT Inhibitor Enzymology

Updated 4 min read

On this pageWhat Is 5-Amino-1MQ?

5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). This reference explains the enzymology of NNMT, how 5-Amino-1MQ interacts with it, how its activity is measured in the laboratory, and how research-grade material is characterized and stored.

What Is 5-Amino-1MQ?

Despite often being listed alongside research peptides, 5-Amino-1MQ is not a peptide. It is a small aromatic cation built on a quinolinium scaffold: a quinoline ring that is methylated on the ring nitrogen (N1) and carries an amino group at position 5. The permanently charged N-methylquinolinium core resembles the product of the NNMT reaction, 1-methylnicotinamide, which is the structural basis for its use as an NNMT inhibitor.

  • Name: 5-Amino-1MQ (5-amino-1-methylquinolinium)
  • Cation formula: C10H11N2+, supplied as a salt with a counter-ion
  • Class: quinolinium small molecule; NNMT inhibitor
  • Enzyme target: nicotinamide N-methyltransferase (NNMT, EC 2.1.1.1)

Because it is supplied as a salt, the counter-ion should be taken into account when calculating molar concentrations from the mass on a certificate of analysis.

NNMT Enzymology: The Reaction 5-Amino-1MQ Inhibits

NNMT is a cytosolic methyltransferase that uses S-adenosyl-L-methionine (SAM) as the methyl donor. It transfers a methyl group to the pyridine nitrogen of nicotinamide:

Nicotinamide + SAM → 1-methylnicotinamide (1-MNA) + S-adenosyl-L-homocysteine (SAH)

Two consequences of this reaction make NNMT a frequent subject of cell biology research:

  • Methyl-donor balance: each turnover consumes SAM and produces SAH, so NNMT activity influences the cellular SAM/SAH ratio (the “methylation potential”) that other methyltransferases, including histone and DNA methyltransferases, depend on.
  • Nicotinamide flux: nicotinamide is the substrate of the NAD+ salvage pathway enzyme NAMPT. Methylation by NNMT diverts nicotinamide into 1-MNA, which is then further oxidized by aldehyde oxidase to pyridones.

Structurally, NNMT is a class I (Rossmann-fold) methyltransferase with adjacent binding sites for SAM and nicotinamide. Kinetic and structural studies of the enzyme describe an ordered mechanism in which SAM binds before nicotinamide.

How 5-Amino-1MQ Works at the Enzyme Level

5-Amino-1MQ belongs to a series of N-methylquinolinium analogs developed through structure–activity work on NNMT. These compounds are understood to occupy the nicotinamide-binding pocket, mimicking the methylated product and competing with the substrate. The aromatic quinolinium ring sits within the hydrophobic substrate site, while the ring substituents influence potency and selectivity.

In research settings, 5-Amino-1MQ is used to ask molecular questions such as:

  • How does NNMT inhibition change intracellular 1-MNA, SAM and SAH levels?
  • Does blocking nicotinamide methylation alter the nicotinamide pool available to the NAD+ salvage pathway?
  • How do changes in methylation potential affect histone methylation marks and gene expression in cultured cells?
  • How selective is the compound against other SAM-dependent methyltransferases?

Measuring NNMT Inhibition by 5-Amino-1MQ

Common assay formats for NNMT and its inhibitors include:

  • LC-MS/MS quantification of 1-MNA formed from nicotinamide by recombinant enzyme or cell lysates
  • SAH-coupled assays, in which SAH is enzymatically converted to a fluorescent or luminescent signal (for example via SAH hydrolase and thiol detection)
  • Cellular target engagement, measuring intracellular 1-MNA and the SAM/SAH ratio after compound exposure in cell lines with known NNMT expression
  • Selectivity screening against related methyltransferases such as INMT, PNMT and other SAM-dependent enzymes

Typical outputs are IC50 values, mode-of-inhibition analysis by varying substrate and SAM concentrations, and concentration–response data for intracellular metabolites.

Characterization of 5-Amino-1MQ Research Material

As a small molecule, 5-Amino-1MQ is characterized with standard analytical chemistry methods:

  • Identity by mass spectrometry (observing the cation mass) and, where provided, 1H NMR
  • Purity by reversed-phase high-performance liquid chromatography (HPLC) with UV detection
  • Appearance and counter-ion statement, so that molar concentrations can be calculated correctly

A certificate of analysis should link these results to a specific lot. For a primer on chromatographic purity, see HPLC testing.

Storage and Handling of 5-Amino-1MQ

  • Store the solid sealed, dry and protected from light; −20 °C is a common choice for long-term storage
  • Allow the container to equilibrate to room temperature before opening to avoid moisture uptake
  • Prepare stock solutions in a solvent compatible with the assay, aliquot them and avoid repeated freeze–thaw
  • Record lot number and counter-ion in laboratory notes for reproducibility

See storage best practices for general guidance. Research-grade material is listed as 5-Amino-1MQ.

NuRev supplies 5-Amino-1MQ for laboratory research use only; it is not for human or veterinary use.
