Melanotan I (afamelanotide) is a synthetic linear tridecapeptide analog of alpha-melanocyte-stimulating hormone (α-MSH), [Nle4, D-Phe7]-α-MSH: Ac-Ser-Tyr-Ser-Nle-Glu-His-D-Phe-Arg-Trp-Gly-Lys-Pro-Val-NH2. The norleucine and D-phenylalanine substitutions make it more resistant to enzymatic degradation than native α-MSH. It is an agonist of the melanocortin 1 receptor (MC1R). In laboratory research it is used to study melanocortin receptor pharmacology and cAMP signaling in cell models.
